Assembly Designs

Assembly Designs will be saved in the "Molecular Biology" Toolkit.

If you want to import a new Design, open your Designs library, click on New Design, and load the Combinatorial Golden Gate design.

You will be brought to the Design editor with the example design you just loaded open for editing. Since this design is ready to submit, let's do so by simply clicking the ‘Submit for Assembly’ button on the upper right.

Once submitted, the design will be sent to the assembler and after a short time, an assembly report is generated with the key information needed to do the assembly. In this case, the assembly will rely on generating DNA assembly fragments via PCR, so detailed PCR information is included in the output report.


Submitting a Design for Assembly

When the report is ready to view, you will see an alert. You can also check the submission's progress via the "Tasks" monitor. To view the report, click on the appropriate row in the Tasks monitor report, the Notifications report, or the Tasks in the left menu.


DNA Assembly Reports Library

You can access directly to them in the "Molecular Biology" toolkit.


To view a construct, double-click on a row in the ‘Assembled Constructs’ table. We will use this assembly report from Design to inform our workflow run. The assembly report contains instructions, such as which materials are necessary for PCR reactions and assembly reactions, leading to the target constructs.


Link Sequences to Materials

DNA Assembly reports sequences can be linked to your "Material Registry" toolkit entities. These sequences will appear in their respective libraries, and you will be able to see their availability in inventory as a material.

Now "Construct_0002" from the DNA Assembly Report above is linked to pre-existent "DNA Materials."

After clicking on the "Constuct__0002" link of DNA Material, you'll be sent to the "Material Registry" toolkit in its DNA Material library.

Now you can see its material lineage, as well as associated samples and aliquots.
